Effect of Hemodialysis Membranes on beta2-Microglobulin Production by Peripheral Blood Mononuclear Cells

Abstract
To evaluate the effect of different dialysis membranes on the production of ¥â2-microglobulin (¥â2m) by peripheral blood mononuclear cells (PBMC0, we measured the ¥â2m concentrations in the supernatant of 24-hours cultured PBMC pre-and post
dialysis
isolated from 9 chronic hemodialysis patients, who have used PMMA membrane over 6 monlths, using the following criteria: (1) in last dialysis with PMMA membranes (3) (PMMA1) (2) in 6th times of dialysis with Cuprophan membranes 93) in further 6th
times
of dialysis with PMMA membranes (PMMA2). And we also investigated the effect of adding PMMA or Cuprophan membrane on the ¥â2m concentration in the supernatant of 24-hours cultured PBMC pre-PMMA1, pre-Cuprophan isolated from chronic hemodialyzed
patients
and normal control subjects.
@ES The results were as follows:
@EN 1) There were no significant difference in ¥â2m concentration of PBMC culture supernatant among normal controls, chronic renal failure patients and chronic hemodialysis patients.
2) There were no significant changes in ¥â2m concentration of PBMC culture superatant according to cross over use of dialysis membrane as well as between pre-dialysis and post-dialysis.
3) In PMMA group, ¥â2m concentration of PBMC cu lture supernatant with PMMA membrane particle (35.2¡¾2.1ng/ml) was low compared to those of PBMC with Cuprophan membrane particle (46.7¡¾4.5eng/ml; p<0.0) and spontaneous PBMC (54.9¡¾3.7 ng/ml:
p<0.001).
4) In Cuprophan group, ¥â2m concentrations of PBMC cultures supernatant with PMMA membrane particle (31.1¡¾1.4 ng/ml( and Cuprophan membrane particle (36.1¡¾2.2 ng/ml) were low compared to that of spontaneous PBMC (46.1¡¾1.9 ng/ml( both
p<0.001).
5) In normal group, ¥â2m concentration PBMC culture supernatant with PMMA membrane particle (34.9¡¾1.9 ng/ml) was low compared to those of PBMC with Cuprophan membrane particle (39.4¡¾1.9 ng/ml:p<0.01) and spontaneous PBMC (44.0¡¾3.6
ng/ml:p<0.01).
These data suggest that PBMC from chromic hemodialysis patients with PMMA membrane behave a normal in vitro ¥â2m production and the direct interaction between PBMC an diasma ¥â2m concentration observed in chronic hemodialysis patients. However,
it
seems
that PMMA membrane has adsorptive capacity for ¥â2m or inhibits the production of ¥â2m by PBMC directly more than Cuprophan membrane.
